Police have used ammunition to disperse protesters in Mandalay, local media and medical staff have reported.
Hundreds of people have gathered in March in Myanmar's second largest city.
Some people are reportedly injured by fire and rubber bullets.
Myanmar is facing protests now and a few weeks, after the pull that has resulted in the departure of the Aung San Suu Kyia government.
Protesters are demanding her release, just like some members of the National League for Democracy.
It is under house arrest, having been charged with possession of illegal radiomakers and violations of the Natural Disaster Law.
The military claims that her party's victory last year has been manipulated, though military officials have not provided data on these claims.
Violence in Mandalay comes a day after a young woman has become the first victim in the protest.
My Thwe Thwe Khaing is shot to death, until human rights groups believe her wounds are caused by ammunition.
Police, on the other hand, have denied using such kind of force.
