The dismissal is part of a cabinet reshuffle, following protests on police brutality.
Castaner has been criticised for tough police actions against protesters, who were prompted by African George Floyd's death during police arrest in Minneapolis, U.S.A.
Macron appointed former Budget Minister Gerald Darman instead.
He also replaced the Minister of Justice and Ecology.
The changes come days after Macro fired Prime Minister Edouard Philippe, after three years in the post, replacing him with Jean Castex.
