A taxi hit the car of Prince Norodom Ranaridddh, 74, and Ouk Phalla, 39, who was traveling with him.
The couple were immediately sent to the hospital, but the prince's wife died a few hours later.
They were traveling to meet political supporters when the accident occurred.
Prince Ranariddhh, half brother of King Nordom Sihamon, heads a political party that is opposing general elections expected to be held next month.
Last year, the opposition party, Cambodia's National Rescue Party, was disbanded and its leader was charged with treason.
