Regional police head Volodymyr Timosko told BBC that more than 400 bodies are thought to be buried there.
“Russia leaves death everywhere”, the Ukrainian president said, citing other mass graves that were reported after the withdrawal of Russian troops.
Ukraine and its Western Alat accuse Russian troops of war crimes. Moscow denies these accusations.
It's still unclear what had happened to the victims, but according to some confessions, many of them may have died of bombings and lack of access to health centres.
There are also signs that some of the tombs may belong to Ukrainian soldiers.
Tymokko said the exhumation will begin Friday to get a clearer view of the number of victims in the town that has been under Russian occupation for more than five months.
In his address Thursday, President Zelensky said that “a clearer and verified” will be presented Friday to the Ukrainian people and foreign journalists.
We want the world to know what's really happening and what the Russian invasion has led us to. Bucha, Marioupol and now, unfortunately, Izyum”, he said.
Ukrainian leader was referring to mass graves that were found in the spring in Bucha, near the Kyiv capital, and also near Marioupolis -- the main Ukrainian port in the south-East now occupied by Russian troops.
Andriy Yermak, head of President Zelensky's office, posted a photograph on Twitter of the alleged mass grave, which also said more information is expected Friday.
A mass show was found in Izyum, Kharkiv region. Black procedure has already begun. All bodies will be launched and sent for forensic examination. You're getting more information tomorrow.

Much of the city's Izyum has turned into rubble, with a local politician telling reporters that more than 80% of the city's infrastructure has been destroyed and that there are still bodies being discovered under ruins.
Izuym and a number of cities in the Kharkiv region were released earlier this month following a recent Ukraine counterphiensy that apparently caught Russian troops by surprise leaving them unprepared to defend their positions.
Ukraine says it has identified more than 21,000 possible war crimes -- including killings of civilians and rapes -- committed by Russian troops since President Vladimir Putin ordered the country's full-scale invasion on February 24th. /Periscopi/
