The Peruth Bar plant, which produces Grad and Smerch weapons for Russian troops, was reportedly involved in flames.
Video footage showed a cloud of smoke rising at a distance, with witnesses claiming to have seen the explosion at the moment of the attack.
Two of those killed were allegedly burned alive, while others allegedly died and buried under ruins.
The plant also produced motor loads for aircraft missiles, navigation missile reinforcements and products for the A-135 missile defence system.
Reports suggested there was an explosion and fire Saturday around 8:00 in the factory sector.
The blast came amid suspicions of sabotage related to a series of fires in key Russian countries during the war.
According to reports, the Russian Investigative Committee is conducting an investigation into Saturday's blast.
Last month the Perm munitions plant announced it had recruited 350 additional staff to boost Putin's war efforts.
The additional workforce would allow the plant to work in three shifts over a 24-hour period to provide ammunition for the war in Ukraine.
This follows reports that Russia has had a lack of missiles aimed at Ukraine.
An earlier explosion at the same defense plant in Perm occurred on May 1, in which three women were killed.
